Abstract
Sepsis kills 11 million people annually, with mortality strongly correlated with time-to-antibiotic administration. Existing diagnostic methods require laboratory infrastructure and 12-48 hours for culture results. We present a multiplexed electrochemical biosensor array for simultaneous quantification of five sepsis biomarkers (procalcitonin, IL-6, lactate, CRP, and presepsin) from a 100 μL whole blood sample. The platform integrates aptamer-functionalized electrodes with microfluidic sample processing, achieving detection limits of 0.01-0.1 ng/mL for protein markers. Clinical evaluation in 187 ICU patients demonstrates 94.3% sensitivity and 91.7% specificity for sepsis diagnosis at 30-minute time-to-result, compared to SOFA score (sensitivity: 74.1%) and conventional culture.
